Unveiling Cellular Interactions: A Mass Spectrometry Approach to Spatial Proteomics

Join us on Tuesday, March 19 at 12:15 PM PT, for a workshop at MSACL hosted by Ionpath. During this, we will cover how MIBI can be leveraged to identify proteomic signatures associated with neurodegenerative resilience and our MIBI platform from instrument to analysis.

We will have an engaging discussion on spatial proteomics and discover how MIBI is reshaping the landscape of cellular interaction analysis. Explore the possibilities, learn from real-world successes, and experience firsthand the power of MIBIsight in unlocking the secrets of the tissue microenvironment.

Proteo-spatial signatures of human resilience to Alzheimer’s Disease captured through multiplexed ion beam imaging

Neurodegenerative diseases, such as Alzheimer’s and Parkinson’s, are characterized by aggregation of misfolded proteins and the progressive loss of neurons and synapses in the brain. However, some individuals show cellular and symptomatic resilience to these diseases despite accumulating a buildup of protein aggregates. In this study, we aimed to identify the local proteomic signatures associated with human neurodegenerative resilience through multiplexed ion beam imaging (MIBI). We analyzed FFPE human brain tissue samples from individuals with Alzheimer’s Disease, those who were cognitively healthy and without protein aggregation, and those who were resilient to the development of Alzheimer’s Disease symptoms in the face of existing protein aggregation. By using MIBI we simultaneously measured the expression of 40 proteins in individual cells and tissues across multiple brain regions in 31 patients. Using cell segmentation, pixel clustering, and spatial enrichment analysis, we aim to find proteomic signatures associated with resilience that are distinct from those of the disease states. Our dataset will provide insights into the underlying cellular and neighborhood phenotypes of neurodegenerative resilience and may inform the development of novel therapeutic strategies for Alzheimer’s Disease.

Multiplexed ion beam imaging (MIBI) has been developed to overcome many of the limitations of traditional proteomic imaging platforms for analyzing the tissue microenvironment. MIBI achieves true subcellular spatial resolution in combination with 40-plex labeling which can be imaged simultaneously to a deep understanding of the tissue microenvironment. With the launch of MIBIplus, Ionpath can take researchers from samples to answers in 6 weeks or less by enabling access to Ionpath’s expertise in sample preparation, imaging, and bioinformatics analysis. Each MIBIplus project includes a comprehensive report of the tissue microenvironment, including population statistics and distance metrics across 50+ cell types, as well as comparisons between sample groupings, such as responder and non-responder groups. Access to MIBIsight is included for MIBIplus customers, giving researchers access to interactive bioinformatics plots and control over the presented results without needing access to a dedicated bioinformatics team.
